Você está doente e cansado de ficar doente e cansado? Uma equipe de pesquisa liderada pelo UNLV está explorando se a razão pela qual às vezes nos sentimos mal é porque as células do nosso corpo sofrem com o lixo que se acumula dentro delas.
Gary Kleiger, professor e chefe do departamento de química e bioquímica da UNLV, juntamente com Brenda Schulman, diretora do Instituto Max Planck de Bioquímica, com sede em Munique, e suas equipes estão trabalhando em maneiras de ajudar nossos corpos a caçar e destruir doenças- causando proteínas.
Eles são os autores de um novo estudo inovador, "As ligases Cullin-RING têm como alvo substratos com parceiros catalíticos geometricamente otimizados", publicado na revista Molecular Cell, que amplia nossa compreensão de como enzimas chamadas ligases cullin-RING (ou CRLs) ajudam as células a obter livrar-se de proteínas que não são mais necessárias. Os resultados também apontam para um potencial calcanhar de Aquiles para proteínas que nos deixam doentes.
"As ligases Cullin-RING (CRLs) são nanomáquinas complexas que são cruciais para os intrincados sistemas de descarte e reciclagem das células", disse Schulman. "As CRLs marcam proteínas defeituosas, tóxicas ou supérfluas com uma pequena proteína chamada ubiquitina, e mutações ou disfunções que prejudicam as CRLs são frequentemente associadas a doenças, como distúrbios de desenvolvimento ou câncer."
A equipa de investigação argumenta que, como as LCR têm funções fundamentais na manutenção do bem-estar das nossas células, é de fundamental importância definir e compreender os seus mecanismos moleculares.
"Este é um trabalho extremamente importante. Por exemplo, durante os confinamentos devido à COVID-19, alguns podem lembrar-se que o lixo se acumulou nas calçadas de pelo menos algumas cidades, causando uma crise de saúde", diz Kleiger. “O mesmo se aplica às nossas células, e doenças humanas podem ocorrer, pelo menos em parte, devido ao mau funcionamento dos nossos sistemas de eliminação”.
Saber mais sobre como as células estão usando esses processos para eliminar materiais desnecessários ou indesejados pode potencialmente acelerar os estudos de descoberta de medicamentos.
"Os investigadores já fizeram progressos surpreendentes no sentido de 'sequestrar' CRLs para caçar e destruir proteínas causadoras de doenças. Muitos acreditam que esta é a próxima grande inovação na descoberta de medicamentos", diz Kleiger. “E ao descobrir ainda mais como as LCR funcionam nas nossas células, os cientistas podem empregar este conhecimento para compreender melhor as doenças humanas e ajudar esta nova plataforma de descoberta de medicamentos”.
As CRLs funcionam marcando proteínas destinadas à destruição com uma pequena proteína chamada ubiquitina, e o laboratório de Kleiger foi o primeiro a mostrar que a taxa de transferência de ubiquitina se aproxima de velocidades que rivalizam com algumas das reações mais rápidas conhecidas na biologia. Os resultados do trabalho atual mostram-nos ainda como as células são capazes de ser seletivas sobre aquilo de que se livram, e as proteínas não são destruídas por acidente.
Nos últimos 20 anos, milhares de cientistas gastaram milhares de milhões de dólares na investigação de um conceito terapêutico para tratar doenças humanas, casando enzimas que participam na destruição de proteínas com proteínas que causam doenças.
As LCR são a escolha esmagadora dos especialistas em descoberta de medicamentos para promover a destruição de proteínas causadoras de doenças, e os medicamentos dependentes de LCR já estão na clínica e podem obter a aprovação da FDA dentro de um a dois anos. O trabalho de Kleiger, Schulman e seus colegas impulsiona o progresso em direção a soluções tangíveis.
O novo estudo é apenas o mais recente dos laboratórios Kleiger e Schulman a mostrar instantâneos das LCRs durante o processo de marcação de proteínas para destruição.
