Investigadores do UCLA Health Jonsson Comprehensive Cancer Center identificaram uma proteína chamada UCHL1 em carcinomas neuroendócrinos altamente agressivos e neuroblastoma que poderia potencialmente ser usada como um biomarcador molecular para diagnosticar esses cânceres e prever e monitorar respostas à terapia.
A equipe também descobriu que o uso de um inibidor de UCHL1, isoladamente ou em combinação com quimioterapia, atrasou significativamente o crescimento e a disseminação de carcinomas neuroendócrinos e neuroblastoma em modelos pré-clínicos.
Suas descobertas foram publicadas na revista Cell Reports Medicine.

Os carcinomas neuroendócrinos, como o câncer de próstata neuroendócrino e o câncer de pulmão de pequenas células, começam nas células que liberam hormônios no corpo e podem se desenvolver em diferentes órgãos, incluindo a próstata e o pulmão. Embora não sejam o tipo mais comum de câncer nesses órgãos, os cânceres desse tipo costumam ter um prognóstico ruim e opções terapêuticas limitadas. As terapias atuais para esses tipos de câncer incluem combinações de quimioterapia, radiação e imunoterapia.
O neuroblastoma é um tipo de câncer que ocorre mais comumente em crianças pequenas e se desenvolve a partir de células nervosas imaturas. Ocorre frequentemente nas glândulas supra-renais, mas também pode se desenvolver no tecido nervoso ao longo da coluna, tórax, abdômen ou pelve. No entanto, dependendo do estágio e dos grupos de risco, esses tratamentos apenas prolongam a sobrevida dos pacientes por alguns meses, enfatizando a necessidade de melhores alvos terapêuticos e abordagens minimamente invasivas para o diagnóstico dessas malignidades.
Para identificar alvos drogáveis para carcinomas neuroendócrinos e neuroblastoma, os pesquisadores primeiro analisaram dados proteômicos disponíveis publicamente e identificaram UCHL1 como uma das principais proteínas drogáveis. A equipe investigou então os níveis de UCHL1 em tecidos de pacientes com vários tipos de carcinomas neuroendócrinos e encontrou níveis elevados de UCHL1 em ??câncer de próstata neuroendócrino, carcinoide pulmonar, câncer de pulmão de pequenas células, neuroblastoma e outras neoplasias neuroendócrinas.
Isto sugeriu que o UCHL1 pode ser um alvo comum para o desenvolvimento de medicamentos em cancros neuroendócrinos com base na sua maior expressão nestes tumores em comparação com tecidos não neuroendócrinos. Esta pesquisa pode ajudar a orientar o desenvolvimento de novos testes de sangue minimamente invasivos para detectar e monitorar respostas a terapias em pacientes com carcinomas neuroendócrinos, como câncer de próstata neuroendócrino altamente agressivo e câncer de pulmão de pequenas células, bem como neuroblastoma.
